【背景知识】

Indoleamine-2,3-dioxygenase (IDO) 催化 L-色氨酸 (TRP) 分解为 L-犬尿氨酸 (KYN)，是该途径中的限速酶。 IDO 活性被认为是先天性和适应性免疫系统的重要调节剂。 它在免疫系统的精细调节中发挥重要作用，例如在肿瘤疾病的发展或过程中。

经典概念是肿瘤或骨髓细胞在肿瘤微环境或淋巴结中产生大量的吲哚胺-2,3-双加氧酶 1 (IDO1)。 这导致局部微环境中的色氨酸耗尽，从而抑制细胞毒性 T 细胞的免疫反应。 同时，由此产生的犬尿氨酸增加了调节T细胞[1]的活性，从而使肿瘤可以逃避免疫系统。

在预防方面，增加 IDO 活性表明癌症风险增加[2]。IDO 在大多数肿瘤组织中也被激活，并且是肿瘤免疫防御的重要因素[3,4]， IDO 活性增加导致 KYN 水平升高与各种恶性肿瘤有关与较差的治疗预后相关，如结直肠癌[5]、肺癌[6,7]、白血病 R、霍奇金淋巴瘤[9]和宫颈癌[10]。

近年来已经开发出干扰这种代谢途径的药物，特别是在 ITE。 这些药物的主要目的是逆转癌症引起的免疫抑制，并且已经在临床试验中[11]。

使用这种 ELISA，可以同时测量一个样品中的 L-色氨酸和 L-犬尿氨酸。 IDO 活动可以通过 KYN/TRP 比率来确定。
